FK506

Calcineurine-remmer

FK506 (Tacrolimus) wordt algemeen gebruikt als immunosuppressivum om de afstoting van orgaantransplantaties te voorkomen.

Gelijk aan cyclosporine A (CsA) blokkeert FK506 de activering van calcineurine door de vorming van complexen met immunofilines. FK506 bindt aan een ander immunofiline dan CsA, genaamd FK506 bindend eiwit (FKBP) 12 .

Verder is aangetoond dat het FK506-FKBP complex de activering onderdrukt van de calcineurine-afhankelijke NFAT-route en de calcineurine-onafhankelijke activeringsroute voor JNK en p38 .
